Only direct upgrades from version 12.0.1 will be possible. If you wish to upgrade to 12.1 from any other version you will need to, upgrade all previous releases first.
I would like to add here that direct upgrades from version 12.0 will also be supported as the only difference in 12.0 and 12.0.1 is just the application wars. The product version in Add/Remove Program is shown as 12.0 even when you upgrade to 12.0.1 patch.
